I tried to build it in a size 5 city where I had yet to build a Cathedral. I got a "bloop" error sound and the click would not stick.
I tried it in one of my "core" cities which has a Cath and it worked!

Shakespeares Lab (seemingly) can only be built in your Capitol!! (I guess it's a "royal theater?")
WHY DON'T THEY PUT THESE STIPULATIONS IN THE MANUAL??!?!
