My stupidest mistake was probably screwing a test question because I had counted 2*3=8 and based my calculations on that. (Happened in high school.)

Being a lib arts major, I can't recall ever having a blatantly stupid (er, sorry Eli) mistake of this sort that screwed me over. My best friend in highschool, however, got serious points taken off a paper he did on the French Revolution because he--consistently--misspelled the word "public" as "pubic."

Gotta love that Committee of Pubic Safety...

I remember the time on my GCSE English mock that I accused Paul Theroux of lying when he said he didn't go into a gypsy camp in London because he resisted the temptation to travel within his own country. I pointed out, in no uncertain terms, that he avoided them because they were a bunch of smelly gypos.

Sort of. But the link was extremely tenuous, IIRC. It was 3-4 years ago, so I don't remember the exact question we were asked.

What I do remember is being stupid on the next question, where I spent 40 minutes criticising the question itself before actually doing it. Once again, the English department went apeshit (quite rightly as well; smart-arsed little **** that I was).
